چکیده

The preparation of a cell-free amino acid incorporation system from Bacillus stearothermophilus active at 65” and the conditions for optimal activity with synthetic polynucleotide messengers are described. The activity of this system is comparable to that of the conventional Escherichia coli system. Its thermal stability, high activity, and low nuclease content are convenient features for studies of protein synthesis and coding. Thus, a 1.4: 1 copolymer of adenylic and uridylic acids, which, because of a high degree of ordered structure, is almost inactive in the E. co2i system at 3’7’, promoted significant incorporation of isaleucine, leucine, and lysine in the B. stearothermophilus system at 65“.

Incorporation of adenine nucleotide into ribonucleic acid by cytoplasmic enzyme preparations of chick embryos.

The incorporation of labeled precursors into ribonucleic acid with cell-free preparations of animal tissues has been observed in a number of laboratories (l-lo). Further studies with partially purified preparations indicate that the proximate precursors are nucleoside trirather than diphosphates (24). These enzyme preparations, unlike deoxyribonucleic acid polymerase (11-13) or polynucleotide p...

On the Significance of Measurements of Amino Acid Incorporation into the Proteins of Cell-free Preparations.

pea seedlings incorporate a variety of C14-amino acids into their protein. The incorporation is promoted by adenosine triphosphate, by a mixture of amino acids, by magnesium and potassium ions (5), and by the nucleosides derived from ribonucleic acid (6). Amino acid incorporation is inhibited by adenosine diphosphate, by amino acid analogues, by rubidium ions, by certain purine analogues, and b...

Naphthalene degradation and incorporation of naphthalene-derived carbon into biomass by the thermophile Bacillus thermoleovorans.

The thermophilic aerobic bacterium Bacillus thermoleovorans Hamburg 2 grows at 60 degrees C on naphthalene as the sole source of carbon and energy. In batch cultures, an effective substrate degradation was observed. The carbon balance, including naphthalene, metabolites, biomass, and CO(2), was determined by the application of [1-(13)C]naphthalene. The incorporation of naphthalene-derived carbo...

Synthesis of hemoglobin in a cell-free system. I. Properties of the complete system.

The incorporation of Ci4-amino acids into protein, with cellfree systems containing ribosomes and soluble enzymes, has been studied extensively since the first reports by Zamecnik and Keller (1). This topic has been reviewed in detail by Hoagland (2). One limitation with many of the cell-free systems has been the failure to find incorporation into known proteins. Incorporation of CY-amino acids...

Molecular characterisation of a DNA ligase gene of the extremely thermophilic archaeon Desulfurolobus ambivalens shows close phylogenetic relationship to eukaryotic ligases.

A 3382 bp fragment containing a gene for a DNA ligase from the extremely thermophilic, acidophilic, and facultatively anaerobic archaeon (archaebacterium) Desulfurolobus ambivalens was cloned and sequenced. The deduced amino acid sequence (600 amino acids, 67619 molecular weight) showed 30-34% sequence identity with the ATP-dependent eucaryal (eukaryotic) DNA ligases of Schizosaccharomyces pomb...
